Creating New Opportunities for Synthesis and Discovery

Nature owes its unparalleled structural and functional diversity to the power of enzymes and multi-enzyme pathways that comprise the synthetic machinery of biological systems. Biocatalysis has generated a vast array of natural products for use as pharmaceuticals and agrochemicals, as well as raw materials for the production of chemical intermediates and polymeric materials. However, only a small portion of Nature’s vast biocatalytic repertoire has been exploited in practice, and a huge assortment of natural compounds and processes, as well as the enzymes and metabolic pathways that generate and empower them, remains largely untapped.

New and high-throughput biocatalytic technologies will be necessary to gain access to Nature’s “warehouse” of structures and functions, and to synthesize new biocatalytic pathways for the generation of novel compounds and materials for use in the pharmaceutical, chemical, and agrochemical industries. Our combined efforts have resulted in the development of several novel techniques that employ biocatalysis, in some cases with high throughput, at both the micro- and nanoscales.

In this joint presentation, we will discuss several approaches that employ biocatalysis as a synthetic tool in high throughput and on very small scales. Small-scale systems include microfluidic reactors in various configurations, and immobilized enzyme microarrays in concert with cell-based screening. Challenges of using enzymes in these platforms in both aqueous and nonaqueous media will be described. Further application of biocatalysis the synthesis of “unnatural” natural products and of nanostructured materials will also be discussed.
